My kitten recently got vaccinated (yesterday) and her stomach is not reacting well. Today she strained to poop, poop was normal brown colour and on the soft side, but not concerning. She meowed a bit while she pooped then threw up afterwards. She went completely back to normal kitten behaviour, running around. Should I be concerned?

As long as she is behaving normally now, there is likely no cause for concern. She may have had a mild vaccine reaction. If she is due for more vaccines, mention this episode to your veterinarian before the next round. Monitor for continued symptoms and see your vet if they persist or worsen.
