I’m doing my first glucose curve at home for my diabetic dog, I noticed when I went to check his blood the second time his puncture from the first blood check started to bleed . Can I use this blood to check his blood the second time or do I need to re- puncture?

If it is a new red blood drop you can use it. That is really not normal though for it to still be bleeding. Keep on eye on that closely. If he keeps bleeding from that spot he needs to be checked by your vet. That should have clotted a long time ago.
