Pet's info: Dog | Mastiff | Female | unspayed | 6 years and 4 months old | 130 lbs
I don't think she's pregnant but when I squeeze on her nipples a dark brown substance comes out, and sometimes it bothers her when I do it. What could cause this? She's 6

She may have some mastitis, an infection in her mammary tissue. I would have your vet take a look at her. Your vet may want to check her white blood count to ensure it isn't elevated. Your vet may also want to put her on some antibiotics. If she is st all lethargic or decreased appetite I would take her in ASAP as an infection can go systemic and cause them to be really ill. I hope this helps and she starts to feel better.
