What's the best way of telling if a female American Staffy is pregnant she was last mated on the 26th of September so if I worked out you should be about 30 something days pregnant. Her nipples are starting to sag. Pet's info: Dog | American Staffordshire Terrier | Female | unspayed | 11 months and 12 days old

The best way would be to have your vet do an ultrasound to look for puppies. Your vet can also let you know how many puppies she is carrying so you know what to expect when she is whelping. If you can't have an ultrasound done than you can continue to look for changes, like nipple growth, behavior changes, enlarged abdomen, and milk production.
